چکیده

Figure 1: Membrane lipids are polar and can be separated into phospholipids and glycolipids, for which the basic structures are shown. Note the difference in the third binding site. Phospholipids have a phosphate bound to an alcohol (glycerol-) or choline (sphingo-), while glycolipids have a sugar group. Source: Lafer, Eileen. “Membrane Lipids I and II: Glycerophospholipids and Sphingolipids.” Lecture. San Antonio, Texas. December 6, 2011 Lipid headgroups are substituents that attach to the glycerol, sphingosine, or cholesterol backbones of polar membrane lipids. Phosphoand glycolipids have headgroups linked to the backbone through a phosphodiester bond (Figure 2). Understanding head group behavior is necessary for the study of membrane biology. The high structural diversity allows for a large range of functions, including effects on membrane curvature [1] cell signaling, substrate transport [2], and more. Consequently, their crucial role in membrane biology also ties them in with a broad range of diseases, from cardiovascular defects to cancer.

Sphingolipid partitioning into ordered domains in cholesterol-free and cholesterol-containing lipid bilayers.

We have used fluorescence-quenching measurements to characterize the partitioning of a variety of indolyl-labeled phospho- and sphingolipids between gel or liquid-ordered and liquid-disordered lipid domains in several types of lipid bilayers where such domains coexist. In both cholesterol-free and cholesterol-containing lipid mixtures, sphingolipids with diverse polar headgroups (ranging from s...

Changes in phosphatidylcholine headgroup tilt and water order induced by monovalent salts: molecular dynamics simulations.

The association between monovalent salts and neutral lipid bilayers is known to influence global bilayer structural properties such as headgroup conformational fluctuations and the dipole potential. The local influence of the ions, however, has been unknown due to limited structural resolution of experimental methods. Molecular dynamics simulations are used here to elucidate local structural re...

Theory of Lipid Polymorphism as a Function of pH

We introduce a microscopic model of a lipid with a charged headgroup and flexible hydrophobic tails, a neutral solvent, and counter ions. Short-ranged interactions between hydrophilic and hydrophobic moieties are included as are the Coulomb interactions between charges. Further, we include a short-ranged interaction between charges and neutral solvent, which mimics the shortranged, thermally av...

The effect of fructan on membrane lipid organization and dynamics in the dry state.

Fructans are a group of fructose-based oligo- and polysaccharides, which appear to be involved in membrane preservation during dehydration by interacting with the membrane lipids. To get further understanding of the protective mechanism, the consequences of the fructan-membrane lipid interaction for the molecular organization and dynamics in the dry state were studied. POPC and DMPC were invest...
